肝星細胞由来の新しい因子の遺伝子クローニングと抗体作製および機能解析

研究概要	肝星細胞(伊東細胞)は特異な機能を有する肝類洞壁構成細胞で、サイトカインや増殖因子などさまざまな因子を産生し、肝疾患の病態に重要な役割を果たしていると考えられている。これまでに形態学的、生化学的およぴ分子生物学的な解析がなされてきたが、その増殖と分化および機能発現にかかわる遺伝子機構はいまだ明らかにされていない。本研究は肝星細胞の分化およぴ機能発現に重要な役割を果たす遺伝子ならびに肝星細胞が有する新しい因子のクローニングおよび抗体作製を目的として施行された。これまでに、SV40温度感受性肝星(伊東)細胞株を大型T抗原が発現する許容温度(33度)およびT抗原が消失して分化が誘導される非許容温度(39度)にて培養し、各々の細胞のRNAから肝星細胞がそれぞれの分化状態において特異的に発現している遺伝子の候補を選択し、クローニングを行った。肝星細胞の静止状態および活性化状態に特異的に発現していると考えられる遺伝子についてそれぞれシークエンスを行い、またこれらの遺伝子の一部については、In situhybridizationおよびnorthern blottingを用いて、静止状態および活性化状態に特異的に発現していることを確認できた。また、この遺伝子を用いて、正常肝組織や肝硬変組織および慢性肝炎組織においてIn situ hybridizationおよびnorthern blottingを行い、肝星細胞およびその活性化がこれらの疾患における肝障害の程度等において重要な役割を果たしているという知見を得た。今年度は、肝星細胞の活性化に伴う細胞内シグナル伝達機構の研究を行い、星細胞の活性化と遺伝子発現に関する論文の投稿中である.クローニングした遺伝子から発現した蛋白に対する抗体作製およびトランスジェニックマウスの作製は続行しており、線維化の遺伝子治療の開発に役立てる目的で研究を行っている。
