マウスにおけるC型肝炎ウイルス(HCV)感染モデルの作製とワクチン効果の判定

研究概要

1.マウスにおけるHCV感染モデルの作製:マウス肝臓に対しHCV構造蛋白のDNAを直接接種し、HCV感染と同様の状態を作製することを試みた。肝臓以外の臓器にDNAが導入されることを防ぐためにDNA接種部位に電気刺激を行い接種したところHCV構造蛋白が免疫組織化学的に認められたが電極間以外では認められなかった。これらDNA接種マウスでは構造蛋白に対し細胞傷害性Tリンパ球(CTL)が誘導されていた。
2.マウスにおけるHCVワクチンの検討(1)ペプタイドワクチン:ペプタイドワクチンによるHCV特異的CTLの誘導を試みた。マウスにCTLエピトープペプタイド免疫ではCTLは誘導されなかったが、Thエピトープを結合させたTh-CTLではCTLが誘導された。さらにCTLエピトープペプタイド単独でCTLを誘導するために細胞質内に直接電気刺激を用いて導入したところCTLが誘導され、さらにImmuno Stimulator Sequence(ISS)、およびB7-1DNAを混合して接種したところ誘導されたCTL活性は増強された。(2))DNAワクチン:elongation factor 1-α(EF1-αプロモーターを用いたプラスミドによりHCVに対するDNAワクチンを試みた。CMVプロモーターを用いた場合CTLの誘導は3回以上の免疫を必要としたがEF1-αプロモーターを用いた場合は1回の免疫で高い活性のCTLが誘導された。(3)リコンビナントワクチン:結核菌ワクチンであるBCGにHCV非構造蛋白NS5のCTLエピトープのみを挿入しマウスに接種したところ高い細胞傷害活性を持つCTLが誘導された。これらワクチン接種マウスにNS遺伝子組み込みワクシニアウイルスをチャレンジしたところコントロールに比べ有意にウイルスを抑制し、高いワクチン効果を示した。
