研究概要

本年度は、研究最終年度にあたり次世代を担う経粘膜免疫の中でも近年注目されている経鼻免疫の誘導メカニズムについて鼻粘膜誘導組織としての鼻粘膜関連リンパ組織(NALT)の役割を中心に検討した。NALTを含めた全身のリンパ節が欠損しているマウス(Id2^<-/->)を用いて解析を行った。Id2^<-/->マウスは、野生型マウスと比較して鼻粘膜および腸管粘膜から産生される総分泌型IgAや血清中IgA量には変化が認められなかったが、血清中IgG量は有意に上昇していた。鼻粘膜固有層中に存在するリンパ球をFACSで解析したところ、CD8^+細胞の著しい減少とNK細胞の欠如が認められた。胸腺依存性(TD)抗原としてOVAと粘膜免疫調節剤としてコレラトキシンを用い、Id2^<-/->マウスに1週間ごとに数回経鼻免疫した。野生型マウスでは3回の免疫で充分な抗体価の上昇が認められたが、Id2^<-/->マウスでは6回目より鼻粘膜洗浄液や唾液中にOVA特異的IgAが検出され、その抗体価は野生型マウスの約10分の1以下であった。しかしながら血清中のOVA特異的IgG抗体価の差は認められなかった。次に、胸腺非依存性(TI)抗原としてLPSをコレラトキシンと共に用い、唾液や鼻粘膜洗浄液中に存在する分泌型IgAに対する反応性を検討したところ、Id2^<-/->マウスでは野生型マウスと同程度の抗体価が検出された。これらの結果をまとめると、NALTはTD抗原に対する抗体誘導には非常に重要であり、TI抗原に対しては必要でないことが示唆された。しかしながらNALTが欠損しているにもかかわらずTD抗原に対して僅かながら粘膜IgA抗体を産生したことは大変興味深い。これらの抗体産生機構がB-1細胞とB-2細胞によって担われているかを現在検討中である。
