IL-7レセプター欠失マウスによる細胞内シグナル分子のT細胞分化における機能解析

研究概要

インターロイキン7(IL-7)はリンパ球前駆細胞の増殖因子であり、生体内におけるリンパ球の分化にきわめて大きな役割を果たしている。本研究の目的は、IL-7レセプター(IL-7R)のシグナル伝達分子のSTAT5によるgermline転写の誘導が、TCRγ遺伝子座のDNA組換え酵素に対するaccessibilityを上昇させV-J組換えを誘導する機構を解明することにある。
まず、IL-7R欠損マウスの胎児肝臓からT前駆細胞を単離し、レトロウイルスを用いてIL-7やSTAT5を導入し、haging drop・胎児胸腺器官培養でT細胞に分化させた。IL-7Rを導入したものでは、コントロールベクターと比較して、細胞数が劇的に増加し、γδT細胞も検出された。一方、活性型STAT5を導入したものでは、IL-7R導入時ほどではないが細胞数がある程度回復した。さらに、その際にγδT細胞の分化が見られた。野性型STAT5を導入したものでは明らかなγδT細胞の分化は認められなかった。また、γδT細胞とαβT細胞の比はIL-7Rを導入した場合よりも活性型STAT5を導入した場合の方が高かった。以上の結果より、活性型STAT5の導入がIL-7R欠損マウスにおけるγδT細胞の分化を回復させたことが示された。また、TCRγ遺伝子のV-J組換えをPCR法にて、germline転写の誘導をRT-PCRにて、組換えシグナルの二重鎖切断端をligation-mediated(LM)PCRにて解析すると、活性型STAT5を導入したものでいずれも強い誘導が認められた。以上のことから、活性型STAT5がin vivoのT細胞分化においてTCRγ遺伝子のgermline転写を誘導し、当該遺伝子座のaccessibilityを上昇させ、V-J組換えを引き起こし、γδT細胞の分化を誘導することが示された。
