T細胞抗原認識における免疫学的分子擬態の解析

研究概要	CD4T細胞による抗原認識はかなり曖昧で、一次構造がまったく似ていないペプチドでも交差反応が成立することがある。HLA-ペプチド複合体としてのvan der Waar's表面、疎水部位や極性の配置、水素結合の供与部や受容部の位置と方向性が一致していれば、交差反応性を示すためである。その意味で、B細胞の抗原認識における分子擬態(molecular mimicry)とT細胞の抗原認識におけるそれは質的に異なる。我々は先ず、15-merのphage random peptide library(20の15乗のdiversityに対してprimary componentが10の8乗しかないlibrary)を用いて研究を開始した。しかし、HLA class IIとrandom peptideをcovalentに結合させたlibraryを用いた研究からも示されたように、特定のT細胞が認識するfull agonistの頻度は予想外に低く(そうでなければすべてのT細胞は負の選択をうけてしまうはず)、発想を変えた方法の開発が必要であった。紆余曲折の末、コンビナトリアルペプチドライブラリーと質量分析を組み合わせることにより、スーパーアゴニスト(T細胞が本来認識する野生型ペプチドよりもはるかに強い抗原活性を有するペプチド)を効率よく同定する方法を開発した。また、特異性未知の1個のT細胞を出発材料としてそのT細胞が認識する野生型ペプチドを同定することに成功した。これらにより、自然界に存在し、なおかつ一次構造上似ても似つかないmimicry peptideを同定することも可能になった。また、擬態分子が誘導する免疫応答を考える際に、HLA class II分子を介して抗原提示細胞側に伝達されるシグナルも重要であることが明らかとなり、これに関しても成果をあげることができた。
