インターフェロンγによるエオタキシン産生抑制-新しい好酸球性炎症の制御機構

研究概要

生体試料測定可能なELISAを確立し、アレルギー性角結膜炎患者の涙液中に(Fukagawa et al.J.Allergy Clin.Immunol.103:1220,1999)、また、喘息患者の誘発痰中には(Yamada et al.Allergy.in press)エオタキシが著増していることを見い出した。ヒト皮膚線維芽細胞、気道上皮細胞株、血管内皮細胞のエオタキシン産生能を比較検討したところ、線維芽細胞が、極めて大量のエオタキシンを産生(Miyamasu et al.Cytokine.11:751,1999)また、線維芽細胞のエオタキシン産生は、Th1サイトカインであるINF-γにより、ほぼ完全に抑制される(Miyamasu et al.Int Immunol.11:1001,1999)ことを報告した。
気道上皮細胞株によるエオタキシン産生は、Th2サイトカインであるにIL-4により増強されINF-γにより抑制されるが、RANTESに関しては、INF-γにより増強される。Th2優位反応であるアレルギー疾患では好酸球走化性ケモカインとしては、RANTESよりエオタキシンが重要であることを示した(Fujisawa et al.J.Allergy Clin.Immunol.105:126-33,2000)。マウス皮膚線維芽細胞の検討でも、エオタキシン産生は、Th2サイトカインであるIL-4により増強され、INF-γにより増強され、INF-γにより抑制されるというヒトと同様の結果を認めた。共同研究を通じて、ヒト肺線維芽細胞(Teran et al.Am.J.Respir.Cell Moll.Biol.20:777-86,1999)、鼻ポリープ(Terada et al.Clin.Exp.Allergy in press.)において同様の結果を認めている。
