全身性自己免疫疾患における組織浸潤T細胞の抗原同定法の確立

研究概要

慢性関節リウマチ(Rheumatoid Arthritis、RA)や全身性エリテマトーデス(systemic lupus erythematosus、SLE)などの膠原病は難治性の全身性炎症性疾患である.RAでは滑膜が増殖しリンパ球が浸潤し、そこには特定のT細胞がオリゴクローナルに集積している。また、SLE患者の末梢血にも強いオリゴクローナルなT細胞の集積が認めらる。このようなT細胞クローンの対応抗原を固定することが病因解明に必要と考えられ、その方法を確立する。そのためにランダムペプチドファージライブラリーをTCRでスクリーニングする方法とを検討した。具体的にはMHCくらすI分子とβー2マイクログロブリンを一本鎖蛋白としてファージ上に発現させ、さらにランダムペプチドを遺伝子レベルで結合させたライブラリーを構築し、T細胞と反応するものを選択してくるという方法である。現在まで、ファージ上にβー2マイクログロブリンとともに発現させたMHCクラスI分子(HLA-B5101)は抗HLAクラスI抗体、抗βー2マイクログロブリン抗体に反応し、弱いながら当該結合モチーフを持ったペプチドを搭載可能との結合も認められた。現在、一旦MHCクラスI分子部分をヒトHLA-B5101からマウスH-Dbに変更し、実際にそのペプチド特異的T細胞に結合できるか否かを検討している。これが確認された段階でランダムペプチドライブラリーの構築に進む予定である。
