パピローマウィルスE6・E7遺伝子導入膵管上皮細胞を用いた膵癌発生機構の解析

研究概要	【研究結果】 (1)単層培養膵管上皮細胞の確立ラット膵管を材料とした膵管上皮細胞の単層培養技術を確立し、さらにヒト膵管を用いた場合の最適培養条件の検討に取り組んだ。ヒト検体の採取機会が少なかったこと、提供されたヒト検体は採取後かなり時間が経過している場合が多かったこと、などから今回の検討ではヒト膵管上皮細胞は単層培養されなかった。今後もヒト検体を用いた検討を継続し、単層培養を確立する予定である。 (2)組換えアデノウイルスベクターの作成細胞死を回避する機能を持つ遺伝子(Bcl-2、Bcl-XL)、細胞増殖を促す働きを持つ遺伝子(膵癌の80%で検出される変異型k-ras遺伝子)を組み込んだアデノウイルスの作成を293細胞を用いたCOS-TPC法にて試みた。 Bcl-XLを組み込んだアデノウイルスについては、細胞に感染させることにより蛋白が発現することをWestern blottingにて確認し、さらにbio-activityをbio-assay(細胞にウイルスを感染させてBcl-XLを発現させることによって、細胞が細胞死を惹起しにくくなることを確認する)にて評価した。具体的には、このウイルスを癌細胞株に感染させることによって抗癌剤シスプラチンに耐性になることを確認した。 Bcl-2および変異型k-ras遺伝子を組み込んだウイルスに関しては、クローンは得られたものの、クローンを293細胞を母地として増殖させてウイルス液にする段階で、タイターが上がらないという現象が起こることが確認された。これらの遺伝子が発現してしまうために、293細胞では十分なウイルス増殖が得られない可能性が考えられることから、CRE-loxP systemなどのconditional expression systemを用いて作成する必要があると考えられた。
