ECL細胞における胃粘膜増殖因子Regの産生調節機構の解明

研究概要

私共は、胃粘膜の増殖因子であるReg蛋白が胃粘膜のECL細胞で産生されていることを明らかとしてきた。本研究では、Reg蛋白がどのような発現調節をうけているかをラットを用いたin vivoとラット胃粘膜より分離したECL細胞を用いたin vitroの検討を行った。in vivoの検討では、胃粘膜損傷のない場合にはガストリンがReg蛋白の発現を促進させていた。一方、胃粘膜に損傷の認められる時にはReg蛋白の発現はガストリンではなくCXCケモカインの一つであるCINC-2βによって促進されていることが明らかとなった。単離ECL細胞を用いた検討では、ガストリンとCINC-2βはそれぞれ独立してReg蛋白の発現を促進することが明らかとなった。また、ECL細胞にはガストリン受容体、CXCケモカインの受容体であるCXCR1が発現していること、胃粘膜の増殖細胞帯の増殖細胞にはReg蛋白の受容体が発現していることも明らかとできた。これらの結果より正常胃粘膜の増殖のコントロールにガストリンが、損傷治癒時の増殖コントロールにはCINC-2βが大きな役割を有しており、これらがECL細胞にそれぞれの受容体を介して作用し、Reg蛋白の産生を促進し、Reg蛋白がその受容体を介して胃粘膜の増殖を促進していた。
