トランスフェリン-遺伝子複合体を用いた進行肝細胞癌に対する血管新生抑制遺伝子治療

研究概要	研究目的:肝細胞癌(HCC)は腫瘍血管が豊富な腫瘍である。この腫瘍血管新生は癌細胞より放出されるvascular endothelial growth factor(VEGF)などの血管新生因子により引き起こされることが明らかになってきた。一方各種血管新生抑制因子も同定され、その遺伝子を用いた遺伝子治療も動物モデルで検討され有効性が示されている。しかしこれらの検討は、アデノウイルスベクターを用いたものであり蛋白発現が一過性で繰り返し投与により強い炎症反応を起こす可能性を含んでいる。また腫瘍への投与は局注であり、全身投与でのtargetingは難しいというのが実際である。抗血管新生療法は腫瘍血管新生を抑制し腫瘍の増殖を抑制する治療方法で、長期にかつ腫瘍局所で高濃度に血管新生阻止因子が存在することが重要である。我々は最近Tf-遺伝子複合体によりin vitroのみならずin vivoにおいても腫瘍特異的に遺伝子導入が可能であることを示してきた。このシステムでは全身投与によっても腫瘍局所への導入が可能であることと、免疫原性がなく繰り返し投与も可能であるという点である。そこで本研究では、Tfとangiostatinあるいはendostatinの遺伝子を同様な方法で結合させた発現ベクターを作製し、進行肝細胞癌モデルマウスに対しその抗腫瘍効果、延命効果および抗腫瘍抗果を検討する。方法と結果:CAG promoterによるangiostatin発現ベクターの作製し、次にTf-angiostatin発現ベクター複合体の作製を当教室で作製した抗Tf-Rモノクローナル抗体(5E7)を固相化したaffinity gel上で行なった。HCC細胞株をTf-angiostatin発現ベクター複合体で処理したところ、遺伝子導入効率は高値でその培養上清により血管内皮細胞株CPAEにapoptosisが誘導された。この様にTf-angiostatin発現ベクター複合体により抗血管新生の可能性が示された。今後endostatin遺伝子あるいはこれら遺伝子のfusion sequenceを用いた発現ベクターを用いて検討を進める予定である。
