Th2細胞への分化にかかわる遺伝子の同定

研究概要	アレルギー反応に重要なCD4+T細胞のTh2への分化過程において高発現している遺伝子を同定すべく、Th2への分化をきたす正常マウスのCD4+T細胞とTh2への分化を全くきたさないSTAT6欠損マウスのCD4+T細胞を取り出し抗CD3抗体及びIL-4刺激後の遺伝子の発現レベルをMicroarrayにて比較したところ、GATA3、CCR4、CCR8などのPositive Control Genesに加えCD25(IL-2 receptor α chain)、Crlr(Calcitonin receptor-like receptor)が正常マウスのCD4+T細胞において高発現していることをみいだした。このうちCD25に関して今年度以下のような現象及び生物学的意義をみいだした。 1.CD4+T細胞を抗CD3抗体にて3日間刺激した後、IL-4を作用させその後のT細胞活性化マーカーの発現を経時的に解析したところ、CD69、CD44などのマーカーはSTAT6非依存性の発現パターンであったのに対し、CD25だけが完全にSTAT6依存性に高い発現レベルが保たれていることをみいだした。 2.上記の高い発現レベルのCD25の役割を明らかにすべく、CD4+T細胞を抗CD3抗体にて3日間刺激した後、IL-4あるいはIL-2にてさらに刺激し、もう一度抗CD3抗体を作用させたところ、CD25の発現の高まった正常マウスのCD4+T細胞はそうでないCD4+T細胞に比しアポトーシスに陥りやすいことを見い出した。このことはSTAT6依存性のCD25の高発現が、過剰に拡大するTh2細胞集団のAICD(Activation-induced Cell Death)によるNegative Selectionに役立っている可能性を示唆している。
