緑膿菌由来の分泌型IL-8誘導因子の精製とその慢性下気道感染症における役割

研究概要	持続性緑膿菌感染症を合併する慢性下気道感染症の患者の気道中には持続的なInterleukin-8(IL-8)産生により過剰な好中球浸潤が認められ、過分泌や進行性の気道障害を招来し、慢性呼吸不全を助長する。本研究では、持続性緑膿菌感染の病態解明を目的として緑膿菌由来の分泌型IL-8産生誘導蛋白質について分離、精製を試み、その生化学的性状について検討した。肺上皮細胞において明らかなIL-8産生誘導を示す緑膿菌5276株の培養上清(d-CSPA)を硫安沈殿で濃縮したSAS-CSPA(>30kDa)をゲル濾過により展開したところ、分画10(分子量32-56kDa)にIL-8産生誘導活性の大半が認められた。この分画の生化学的性状はやや熱処理に弱い蛋白質であった。さらに、SAS-CSPAを連続するイオン交換クロマトグラフィーを用いて展開することで単一の高いIL-8産生誘導活性を得た。この分画の生化学的性状も熱処理にやや弱い蛋白質であった。この分画のSDS-PAGE後の銀染色では推定分子量42kDaの位置に明らかな単一バンドを得た。さらに、この分画のゲル濾過による展開でも単一のIL-8産生誘導ピークを確認した。以上の結果より、肺上皮細胞における緑膿菌培養上清中の主要なIL-8産生誘導因子は推定分子量42kDaの熱処理にやや弱い蛋白質と考えられた。今後、この主要なIL-8産生誘導蛋白質の同定とともにその臨床的意義の解明、さらには肺上皮細胞におけるIL-8遺伝子発現機構の解析が必要である。
