フォンビルブランド因子依存性血小板血栓の阻害物質の分子設計

研究概要	(1)蛇毒:タイコブラ(Naja naja kaouthia)蛇毒からkaouthiagin名づけた新規metalloproteaseを精製し、このプロテアーゼがフォンビルブランド因子(VWF)を特異的に切断するユニークな酵素である事を同定した後、蛋白生化学的手法にて、その全アミノ酸一次構造を決定した。さらに同蛇粗毒からL-amino acid oxidaseを同時に精製し、そのN末端部分アミノ酸配列を同定し、既報のものとは異なる事を確認した上で、アゴニスト惹起正常ヒト血小板凝集、並びにアゴニスト非存在下のずり応力惹起血小板凝集に及ぼす影響を観察し、その特有な阻害効果を確認し、同毒蛇咬傷時の過剰出血の機構を一部明らかにした。 (2)von Willebrand factor-cleaving protease (vWF-CPase):1960年代からその存在が知られるも、病態は不明であった遺伝性出血症、Upshaw-Schulman症候群、がこの酵素活性の先天性欠損症である事を始めて実証した。また、これらの患者に新鮮凍結血漿を輸注(FFP,10ml/kg)すると、血中の酵素活性はほぼ期待値まで上昇するが、輸注2日目には酵素活性はほぼ消失する事、一方、血小板数はこの後急速に増加して、7-l0日目に最大プラトーとなり、その後再び低下していく事を示した。さらに、患者血漿中の超高分子量VWFマルチマー(UL-VWFMs)の出現により、患者血漿と正常ヒト洗滌血小板を混じたずり応力惹起血小板凝集の最大凝集値が著しく亢進するも、FFP輸注により凝集は正常化する事を示し、UL-VWFMsの病態生理学的意義を明らかにした。現在、これら患者と家族のVWF-CPase遺伝子(ADAMTS13)の解析を実施中である。 (3)胎盤由来ecto-ATPDase : Enzyme IとIIと名づけたサイズの異なる二つの胎盤ecto-αATPDaseのcDNAを、それぞれCOS-71細胞で発現し、まず、Enzyme IはCD39とは異なり、天然型の可溶性ecto-ATPDaseである事を確認できた(論文未発表)。
