HHV-6初感染ならびに再活性化の病態解明と制御に関する総合的研究

研究概要

1)高純度なウイルス抗原液作製:in-vivoの実験を実施するにあたり、バックグラウンド値を低下させる必要が生じた。そこで、より純度の高いウイルス抗原液を作製するため、HHV-6感染臍帯血単核球をSPGA mediumに溶解後、ショ糖密度勾配遠心法にてウイルス粒子を精製して抗原液として使用した。その結果、約80%のヒトにおいて、バックグラウンドが減少することが確認された。しかしながら、約20%のヒトでは、相変わらず高いバックグラウンド値が観察された。これは、ウイルス抗原液作製のために使用した臍帯血単核球の保持していたHLA型が影響していると推測された。
2)K562細胞への感染実験:K562細胞は、細胞表面にHLAを発現していないリンパ球系細胞である。上記実験1)にて問題となった高バックグラウンド値の問題を解決するために、HLAの発現していないK562細胞でのウイルス抗原の精製を試みた。高力価のHHV-6液を作製し、K562細胞への感染実験を実施した。MOI=0.1で感染させ、IEA/ex3抗体を用いた間接蛍光抗体法で感染の有無を確認した。Controlとして使用した臍帯血単核球においては、感染後24時間で約80%の細胞が抗原陽性であった。しかしながら、k562細胞については約0.1%の細胞で特異抗原が観察されたのみで、感染効率は極めて悪いことが明らかになった。
3)骨髄移植患者でのリンパ球増殖反応の検討:移植後1ヶ月目の患者、5例について精製したHHV-6抗原を使用して実施した。5例間でばらつきはあったものの、S.I.の平均値は2.8であり以前測定した健康成人の値と比較して低値を示した。
