ヒト1番染色体短腕の1p36領域における神経芽腫関連がん抑制遺伝子の探索

研究概要

ヒト1番染色体p36.2-p36.3は神経芽腫関連領域といわれている。昨年度までの本研究におけるLOH解析の結果、この領域の中でも特にDlS450からDlS244までの約1Mbの領域が可能性が高いという結果が得られた。これに基づき、この領域について神経芽腫関連遺伝子のポジショナルクローニングを行っている。本年度は以下の成果が得られた
1)DlS244周辺領域におけるホモ欠失の同定
上記の領域について40種類の神経芽腫細胞株を用いてホモ欠失の有無を検討したところ、2種類の細胞株にDlS244周辺領域におけるホモ欠失を同定した。そこで、その欠失領域をカバーする約800kbのPACコンティグを作製し、cDNAライブラリースクリーニングとPACクローンDNAのラフシークエンシングにより遺伝子の同定を行った。その結果、6つの遺伝子をこの領域に同定した。これらの遺伝子は代謝酵素PGDH、ソマトスタチン様ペプチドCORTの他、アポトーシスに関連するDFF45、ユビキチンープロテアソーム系に関連するUFD2とPEX14、キネシンスーパーファミリーのひとつKIF1Bbetaであった。(Ohira et al.Oncogene2000)
2)がん抑制遺伝子としての可能性についての検討
上記の6つの遺伝子について神経芽腫におけるがん抑制遺伝子としての可能性について検討した。まず4つの遺伝子について神経芽腫サンプルを用いた遺伝子変異検索をPCR-SSCP法にて行ったが、変異の頻度は非常に低かった。さらに各遺伝子の神経芽腫での発現パターンを見るため、各種神経芽腫細胞株および患者サンプルのRNAを用いたRT-PCRを行った。この時、比較すべき正常組織が入手不可能なため予後良好タイプの神経芽腫と予後不良タイプの神経芽腫とで比較した。少なくとも3つの遺伝子に関して予後不良群における発現が予後良好群に比較して1/3以下に低くなっていたことから、この領域がメチル化などのimprintingを受けている可能性が考えられた。近年いくつかのがん抑制遺伝子ががん組織においてimprintingによる発現低下を引き起こしていることが知られているため、これについては引き続き解析をすすめる必要がある。
