腫瘍の分子病理学的特性と放射線生物学的パラメータの関連性の解明

研究概要	放射線抵抗性の原因の一つである休止期細胞(G0細胞)に着目して、細胞周期をG2/Mでblockするタキサン系新規抗癌剤docetaxelが、細胞周期、特にG0細胞分画に与える影響を検討した。C3Hマウスに移植した直径約10mmのSCCVII腫瘍を実験に用いた。Docetaxel(90mg/kg)の処置後、経時的に腫瘍を切り出しsingle cellに処理し、増殖細胞分画は抗Ki-67抗体で、cell cycleの検討はflowcytometryで測定した。G0細胞分画は、G0/G1期にあり、かつKi-67を発現していない分画として求めた。Docetaxel投与36-72時間では、G1期の細胞がG2/Mに進みG2/M期の比率が最大に達した。このような細胞周期の変動にもかかわらず、G0細胞分画の変動はなく、最大観察期間の投与後120時間までrecruitmentは起きなかった。以上、新規抗癌剤タキソールがG0期細胞からG1期細胞へのrecruitmentを阻害することを明らかにし、本剤が化学放射線療法において腫瘍細胞の加速腫瘍増殖(accelerated repopulation)を抑制しうることが示唆された。 Erythropoietin(EPO)は低酸素により誘導され、赤芽球の分化に関与し、赤血球の産生を増加させる。近年、EPOが血管新生に関与するという報告が見られる。我々はラット化学物質誘発肝癌モデルを用いてEpo,Epo-r,VEGF,flt(VEGF receptor)などの免疫組織化学染色による形態学的解析を行うとともに、それらの蛋白量を定量した。本実験の結果、肝腫瘍からEPO-rが検出されたが、正常肝組織、肝硬変組織からは検出されなかった。また、腫瘍内EPO濃度と血管密度は相関した。肝腫瘍の血管新生にEPOが関与しているものと推定された。
