情動を司る神経回路におけるセロトニン受容体を介した情報伝達の役割に関する研究

研究概要

1.Dex慢性処置後の5-HT-2A受容体および細胞内情報伝達系
(1)DOI誘発性WDSはDex反復処置ラットで有意に亢進していた。
(2)5-HT-2A受容体数はDex反復処置ラットで有意に亢進していた。
(3)phospholipaseC、Gq、inositol-1,4,5-trisphosphate receptor量はDexにより変化しなかった。
(4)KCI刺激性[Ca^<2+>]i変化はDex処置群で亢進していた。
2.5-HT-2A受容体機能に対する開頭とカイニン酸注入の影響
(1)開頭術施行群では、非手術群に比べてWDSが増加していたが、カイニン酸処置群では開頭術によるWDSの増加が回復していた。
(2)5-HT-2A受容体密度は、非手術群、開頭術施行群、カイニン酸投与群の3群で有意な差が見られなかった。
3.自然発症高血圧ラット(SHR)における一過性脳虚血と慢性Dex処置
(1)DOI誘発性WDSはDexのみ処置したラットでは変化なかったが、diazoxide単回処置後にDex反復処置すると、WDSの亢進が観察された。
(2)5-HT-2A受容体数はどの群でも変化なかった。
4.神経系由来培養細胞におけるセロトニン-2A受容体の調節
セロトニン-2A受容体のfull agonistであるセロトニンまたはpartial agonistであるDOIを前処置した後のセロトニン刺激性細胞内カルシウム動員は同程度に低下した。
この時のセロトニン-2A受容体mRNAをPCRで測定したところ、DOI前処置群でのみ増加していた。
