核受容体転写制御におけるポリADPリボースポリメラーゼの役割

研究概要	全長RXRaとGSTの融合蛋白をプローブとして用い、ラツトGH3細胞から構築したcDNA発現ライブフリーをスクリーニングすることによって、生化学的な手法を用いPARPの部分cDNA(a.a82-220)を単離した。ポリADPポリメラーゼ(PARP)は、NAD+を自ら結合し相手の蛋白をポリADPリボシレーションする蛋白修飾酵素であり、その代表的基質は、コアーヒストンを含めたヒストン及びHMG蛋白などクロマチン構築蛋白が有名である。単離したフラグメントは82-220アミノ酸部分でDNA binding domainの二つめのZnフインガーを含む部分であった。PARPがTR-RXRのDNA結合能におよぼす影響をゲルシフト法で観察してみると、PARPは標的DNA配列上に結合したTR-RXRに結合し、さらにシフトした複合体を形成する。COS1細胞を用いたレポーターアッセイでPARPを大量発現させるとT3依存性の転写活性化が著明に低下する。この転写抑制は核受容体に特異的であり、サイトメガロウイルスのプロモーターおよびTPAで刺激されたserum response element(SRE)には全く影響しない。さらに、ポリADPリボシル化酵素活性の欠失変異体PARP(C908R)はT3による転写活性化には影響しないことよりプロモーター上にリクルートされたPARPの酵素活性が重要であるということを示唆している。興味深いことにDNA condensationに重要なヒストン部位が特異的にリボシル化を受け、クロマチン構造とヌクレオゾームのポジショニングがADPリボシレイションの標的であることは転写調節に関係していることを示唆している。DNAに結合した受容体にリクルートしたPARPはヒストンをポリADPリボシル化しアセチル化、脱アセチル化反応を修飾しているものと考えられる。PARPとグルココルチコイドとのキメラ蛋白はグルココルチコイド受容体応答配列に結合し転写を抑制することが示されており、PARPはプロモーターにリクルートされたとき転写抑制因子として働くものと考えられる。
