組換えアデノウイルス系を用いたインスリン感受性PDEキナーゼ活性化機構の解析

研究概要

インスリンはinsulin-sensitive phosphodiesterase(PDE3B)を活性化しcAMPを低下させることにより抗脂肪分解作用を発揮する。申請者らはcell free系でのPDEキナーゼアッセイ系を確立しインスリンにより活性化されるPDEキナーゼはPI3-キナーゼの下流にある未知のキナーゼであることを明らかにした。そこで本研究ではPI3-キナーゼ及びその直下にあると考えられるAktをアデノウイルス系を用いて脂肪細胞に遺伝子導入し、PDEキナーゼ活性化機構におけるこれらの役割を明らかにすることを目的とした。昨年度は神戸大学医学部第2内科小川博士らにより提供された組換えアデノウイルス(dominant negative Akt,dominant negative PI3-K)及びβ-GALを発現するコントロールウイルスをそれぞれ293細胞に感染させ高力価ウイルス液を調整した。そこで本年度はこれらのウイルスを3T3L1脂肪細胞に感染させ、β-GALの発現及びAktまたはまたはPI3-K蛋白の発現をウェスタンブロットにより確認し最適のウイルス力価を算定した。現在これらの組み換えウイルスを3T3L1脂肪細胞に感染させ、インスリンとともにインキュベートし、PDE活性、PDEの燐酸化、PDEキナーゼ等を解析しAktまたはPI3-KがPDEキナーゼ活性化系に関与するかどうか解析を進めているところである。
