細胞周期調節遺伝子発現の解析による食道癌の増殖制御機構の解明

研究概要

G1サイクリンの過剰発現は様々な悪性腫瘍で報告されている。その悪性増殖および予後不良に関する関連はまだ一定の見解を得るに至っていない。今回の研究では、食道癌111例の連続切片を用いて2つの代表的なG1サイクリンであるサイクリンEとサイクリンD1の免疫染色を行った。サイクリンE蛋白の高発現が腫瘍の大きさや深達度によく相関し、結果としてサイクリンE蛋白の高発現の症例は予後不良であることが明かとなった。興味深いことに、Cyclin E蛋白発現の高い細胞は、主に細胞周期がよくまわっている細胞集団で形成されていると考えられる腫瘍の最深部に存在していた。また、PCNA陽性細胞は非常に良くサイクリンE陽性細胞と同部位に存在した。一方、サイクリンD1蛋白は分布に一定の傾向を認めず、腫瘍の増殖や予後と相関を認めなかった。さらに主病巣の時間的に早期の段階を反映していると考えられる、主病巣に併存する粘膜内癌のCyclin Eに関する免疫染色を行った。結果は、併存粘膜内癌がサイクリンE陽性である症例はCyclin E陰性症例に比較し、主病巣が進行していた。これらの結果より、Cyc1in Eの高発現は食道癌により高い悪性度を与え、結果としてより悪性度の高いふるまいをさせる可能性が示唆された。
