肺癌特異的細胞障害性Tリンパ球クローンの誘導および解析

研究概要	本研究は肺癌に対する特異的免疫機構解明の一助とすると共に将来の癌特異的免疫療法の基礎とすることを目的とし、肺癌における腫瘍免疫のメカニズムの研究及び肺癌特異的免疫療法の開発のためのtoolとして肺癌特異的細胞傷害性Tリンパ球クローン(CTL)を誘導し詳細な解析を行なった。肺癌に対する細胞障害性Tリンパ球(CTL)の誘導効率を改善するため株化した肺癌細胞株にCD80遺伝子の導入を行い、CTL樹立を以下のごとく行った。肺癌切除標本550例より肺癌細胞株16株を樹立し、9例中8例でCD80遺伝子のstable transfectionに成功した。CD80が移入された8例全例でCTLが誘導可能であった。限界希釈法により5症例からCTL cloneを樹立し、これらcloneの認識する腫瘍抗原の同定やTCRの解析を進めている。腺癌患者B203においてはHLA-A24拘束性に、A110症例からHLA-Cw12拘束性に自己腫瘍以外にも肺腺癌、肺扁平上皮癌、メラノーマをも傷害するCTL cloneが得られ、癌特異的免疫療法のターゲットになりうる共通抗原の存在を示唆した。ほとんどの症例からCTL cloneが得られたことより、肺癌においても宿主リンパ球に認識されうる腫瘍抗原が存在することが明らかになった。今後これらCTL cloneの認識する腫瘍抗原の同定・解析およびTCR解析は癌特異的免疫療法さらにはTCR特異的PCRを用いた癌特異免疫療法時の免疫学的モニタリングの開発に有用と考えられた。
