虚血性神経組織(Penumbra)のin vivo系におけるモデル化とその応用

研究概要

"虚血性神経半影(Penumbra)のモデル化とその応用"を目的として、in vivo Ishemic Penumbraの応用、今回は「in vivo penumbraモデルにおけるcortical spreading depression(CSD)の発現とcortical spreading depression誘発負荷の影響」を検討した。Wistar ratの右頚動脈を結紮後、hypobaric hypotension techniqueを用い平均血圧を50mmHgに低下させ、かつCSDを誘発した群はCSD後、本来起こるべき脳血流の一過性の増加は認められず、DC potentialは一過性に陰性化した後に正常化した。cortical tissue impedanceは血圧を正常に戻すまで30分間にわたり高値を持続した。組織学的には右大脳皮質のneuron densityはCSDのみを誘発した群、hypotensionのみの群と比較して有意に低下していた。Hypobaric hypotension techniqueにより低潅流域となった脳組織にエネルギー要求性のCSDを誘発したこのmodelはpenumbra-like conditionを再現していると考えられた。
