骨移植における骨髄細胞の骨への分化メカニズムの解析とBMPの骨移植有効化への応用

研究概要	(1)骨(骨髄)移植時の骨形成と同様の過程を示す骨折モデルで、経時的な骨形成因子(BMP-4)とBMPの拮抗因子のひとつであるNogginの発現をnorthern blottingとin situ hybridization法を用いて検討した。その結果、BMP-4とNogginは骨折初期に発現のピークを持ち全過程で同様の局在で共発現していた。 (2)骨(骨髄)移植の実験モデルとしてラット大腿骨骨髄細胞を採取し、これをdiffusion chamberに封入してラット背筋内に埋め込む実験を行った。chamber内の組織の経時的変化は1〜2週でフィルター内側面に細胞成分が付着し3〜4週目に軟骨及び骨形成が進行し、全体として骨折治癒過程での骨折部周辺の変化と同様の現象が見られた。またin situ hybridization法を用いた検討ではフィルター内の細胞の一部はBMP-4のmRNAを発現していた。以上のことから骨髄細胞は骨折や骨移植の際に自身で骨形成因子を発現して分化、増殖し骨形成を誘導しうることが考えられた。 (3)骨吸収促進作用をもつ炎症性サイトカインであるTNF-αに注目し、TNF-αの働きを抑制する薬剤(キサンチン誘導体であるPentoxifylline)の骨量増加作用について検討した。その結果Lipopolysaccharide(LPS)投与によってTNF-α産生を誘発したマウス大腿骨は骨粗鬆化が生じPentoxifyllineの同時投与によりその骨粗鬆化は抑制され、またPentoxifylline単独投与でマウス大腿骨の骨量増加が起こることを証明した。さらにrhBMP-2による異所性骨形成を誘導する実験系にPentoxifyllineを投与してその効果を評価し、PentoxifyllineがrhBMP-2の異所性骨形成作用を増強することを確認した。
