CA125遺伝子プロモーターの同定と卵巣癌特異的遺伝子治療の試み

研究概要	EMBL SP6/T7 phage libraryよりCA125遺伝子のgenomicDNAの上流500bpをprobeにしてクローニングしたgenomicDNAを、exon1よりさらに上流5kbまでシークエンスを行った。CA125遺伝子のexon1は、exon1Aおよびexon1Bよりなっており、正常リンパ球ではexon1Aから、卵巣癌細胞株ではexon1Bからの遺伝子発現が認められるとされていることから、転写開始点をSMART PCR cDNA Synthesis kitにより確認したところ、従来exon1Bに転写開始点のあることが明らかとなった。このため、exon1Bの転写開始点を含めてanti-sense primerを設定し、PCRにより250bpから500bp毎にsense primerを設定することによりdeletion mutantを作成し、dual luciferase assay kit(Promega)を用いることにより、luciferase assayを行いCA125遺伝子の転写活性部位の特定を行った。500bpのところにプロモーター活性を認め、1875bpの所にエンハンサー活性のあることが明らかとなった。卵巣癌細胞株HEY、子宮頚部扁平上皮癌細胞株SKGIIIa、正常ケラチノサイト細胞株K42を用いて、CA125遺伝子プロモーター活性の臓器特異性について検討した。CA125のプロモーター活性は、1875bp上流域に最も高くHEYではSV40プロモーター活性の7倍あり、一方SKGIIIa、K42においてはそれぞれSV40の1.3、1.8倍であった。このことによりCA125遺伝子プロモーターは卵巣癌特異的に非常に高い活性を有することが明かとなった。さらに、conditioned replicative adenovirus vectorを作成するため、E1Aプロモーター部分をCA125遺伝子の上流域1875bpのプロモーターと置換したところHEY細胞においてIC50は0.01MOI、SKGIIIa細胞において100MOIと非常に臓器特異性が強い増殖抑性効果を示した。以上により、CA125遺伝子プロモーターを用いた卵巣癌に対する遺伝子治療は、卵巣癌臓器特異的であり将来的な臨床治療に有用であるものと思われた。
