婦人科癌におけるDNA修復遺伝子、βカテニン、SKT11などの遺伝子異常解析

研究概要	婦人科癌における各種遺伝子異常の解析を行った。まず、ベータカテニン遺伝子の変異、及びその蛋白の発現を卵巣癌と子宮内膜癌の組織切片を用いて研究し、卵巣癌5例(8%)、子宮内膜癌5例(14%)にベータカテニン遺伝子の変異を認めた。また、Peutz-Jegher症候群の原因遺伝子として1998年に報告されたSKT11遺伝子の変異を卵巣癌においてSSCP-Sequence analysisで検討し、exon6のCCG-CTG(Pro281-Leu)の変異を60例中1例で認めた。更にc-srcのactivating mutationが大腸癌で発見されたので、婦人科癌でも検討したところ卵巣癌で0/70、子宮内膜癌で1/68と婦人科癌では低頻度であることがわかった。また、卵巣癌における抗癌剤効果発揮にアポトーシスが関与していて、アポトーシスが、進行卵巣癌患者でその上昇が報告されているインターロイキン-6(IL-6)により、negativeにコントロールされていること、逆に抗IL-6抗体を抗癌剤と併用することによりアポトーシスを介した抗癌剤効果の増強を認めることを細胞株を用いて見出した。そして,卵巣癌細胞株におけるこのIL-6によるアポトーシスを介した抗癌剤効果のコントロールに、抗アポトーシス遺伝子Bcl-2蛋白の量的変化は認めず、Bcl-2を経由しない経路でのアポトーシスのコントロールが示唆された。現在、このアポトーシスをコントロールする各種アポトーシス関連遺伝子の異常と抗癌剤耐性の関連についても検討中である。
