視覚感受性期の前後別の視神経遮断に対する視中枢受容体の変化の研究

研究概要	【目的】感受性期初期の片眼からの入力の途絶が視覚中枢に及ぼす影響を見る目的で、ラット片眼を処置しその脳内変化を検討した。【方法】ウイスターラット110頭を用い、生後10日のラット右眼を眼球摘出または瞼裂縫合し、11Cと14Cをダブルトレーサーとするex-vivo autoradiographyによって、各々1、5、20、55日後の上丘、外側膝状体、視覚領皮質での糖代謝、アデノシンA1受容体密度(節前マーカー)、ベンゾジアゼピン受容体密度(節後マーカー)を測定し、定性的糖代謝率および受容体への特異的結合を左右比で解析した。【結果】眼球摘出ラットの神経活動の指標である糖代謝変化は最大の上丘で処置後1日は5±3%低下、節前マーカーは処置後20日に最大で26±7%の進行性減少、節後マーカーは処置後20日に7±3%の補償性増加(各々n=6)を示し、瞼裂縫合ラットの各指標は有意の変化を示さなかった。【考按】我々の先の実験で成熟ラットは片眼摘出により対側の視覚関連領域の上丘の糖代謝の50%低下、脱神経に伴う節前性受容体密度の50%低下および節後性受容体密度の30%の代償性増加を示した。片眼遮閉した感受性期ラットの視覚領では眼優位性コラムが開放眼側に偏ることから、片眼処置の影響は成熟期ラットよりも化塑性が残る幼若期ラットで更に強いと予想した。しかし感受性期初期の操作では逆に脳の代謝と受容体密度に大きな変化は無く、この結果を説明するには、感受性期における眼障害による視交叉の非交叉性成分の増加などの新たな要素を検討に加える必要があると考えられた。【結論】幼若期ラットでは視覚系に対する片眼入力の遮断が、成熟ラットと異なり左右両側の視中枢に影響する可能性がある。
