研究概要	本研究はPTHおよびそのセカンドメッセンジャーであるcAMPの軟骨培養細胞増殖分化機構を検索する過程で培養液中に蓄積してくる20kdaltonの蛋白質がRBPであることを新たに見いだしたことにその端を発している。これまで軟骨細胞がRBPを生成していることは知られておらず,レチノイン酸の標的器官としての軟骨組織自身が各分化段階でRBPを生成していることの生理的意義を解明することは大変興味深い。本研究は軟骨細胞で生成されているRBPの役割について明らかにすること、そのPTH-cAMP系による生合成調節機構を解明することを目指している。これまで肝臓以外でRBPを生成している臓器としては脳、腎臓、涙腺、脂肪細胞などが知られているがその生成量は肝臓に較べて極めて少ない。その生理的意義については組織へのレチノールの取り込みに機能しているという報告もあるが、まだ明らかではない。これまでの本研究の過程で軟骨細胞は肝臓に次いでRBPの合成量が高く、またPTH/PTHrPなどによっても誘導されることを明らかにしてきた。また、上記の調節因子のうちでPTHが軟骨の最終分化を抑制するのに対してレチノイン酸は最終分化を促進するという互いに異なる方向への作用を及ぼしていることを明らかにしてきた。このような背景の中でPTH-cAMP系がRBPを誘導することの持つ意味を解明することはこれまでになかった視点である。本研究の過程で形態形成因子であるインディアンヘッジホッグの発現がPTH-cAMP経路で著しく抑制され、逆にレチノイン酸で強く誘導される現象が見いだされ、軟骨細胞の増殖・分化過程におけるPTHとレチノイン酸の拮抗的役割の重要な過程がインディアンヘッジホッグ発現への調節を介しても制御されている事が明らかとなった。
