水チャネル蛋白質、アクアポリンの唾液腺細胞における発現と機能調節の分子機構

研究概要	平成11年度に引き続き、12年度も水輸送に関与する蛋白質アクアポリン(AQP)の研究を行った。AQP存在が1992年、Agre等によって明らかにされ、現在まで、多数の新規AQP遺伝子が報告されてきた。1995年にクローニングされた外分泌腺型アクアポリンAQP5はそのペプチド構造中に蛋白質リン酸化酵素Aの標的となる構造が含まれていることから、本チャネル蛋白質はリン酸化され、それにより細胞内の小胞から細胞膜への移動(トラフィッキング)が起こる可能性がある。実際、腎集合管ではAQP2のC末端に近いSer(256)がADH依存性にリン酸化され、このリン酸化がAQP2のトラフィッキングに関与することが示唆されている。11年度は培養唾液腺細胞に遺伝子導入によって発現させたAQP5の動態を観察しそのトラフッキングについて究明した。平成12年度は、大量に水輸送を行っている消化管におけるAQP5の発現、局在、および調節について研究し、以下の結果を得た。(1)AQP5は多くの消化管に発現し、とくに胃と十二指腸においてその発現は強かった。(2)十二指腸に発現しているAQP5のmRNAおよび蛋白質のサイズは唾液腺や肺と同様それぞれ1.9kbp、および27kDであった。(3)免疫組織化学により、十二指腸ではAQP5はブルナー腺の腺腔側膜に発現していた。(4)消化管ホルモン、VIPの刺激は十二指腸AQP5のトラフィッキングを惹起した。(5)ブルナー腺ではAQP5に加えてAQP1も細胞膜に発現することを明らかにし、AchやVIPによってトラフィッキングが起ることを見いだしている(未発表)。これらについて、現在研究続行中である。
