抗酸化剤によるヒト口腔扁平上皮癌細胞死におけるHIF-1αの挙動

研究概要	アスコルビン酸、没食子酸、エピガロカテキンガレート(EGCG)等の抗酸化剤は、ヒト口腔扁平上皮癌HSC-2や、唾液腺腫瘍細胞にDNA断片化(TUNEL法)やカスパーゼの活性化(M30モノクローナル抗体で検出)等のアポトーシスに特徴的な変化を誘導した。また、タンニンの中では、環状構造を持つ加水分解性タンニン、フラボノイド類の中では、イソプレニル化合物が最大活性を誘導した。ポリプレニルアルコールの中では、イソプレニル単位が4個のゲラニルゲラニオールが、また、ビタミンK_2誘導体の中では、イソプレニル単位が2個のMK-2が最大活性を示した。これらの物質は、ヒト歯肉線維芽細胞よりも、口腔癌細胞に対して障害的に作用した。この原因については、今後の解析が待たれる。EGCGを添加することにより、生存シグナル(bcl-2、akt)が減少し、カスパーゼ9及び3の活性化を介して、ヌクレオソーム単位のDNAの断片化が誘導された。アスコルビン酸の添加により、培養液中にラジカル、過酸化水素、メチオニンの酸化物が産生され酸化状態が生じたが、細胞内活性酸素(ROS)の産生は減少した。細胞内外の酸化還元状態の変化については、今後の検討課題である。また、これら抗酸化剤の細胞障害活性は、低酸素誘導因子(HIF1α)を誘導されるとされる塩化コバルトで減弱したが、これは、塩化コバルトによる抗酸化剤の酸化による活性の低下なのか、それとも細胞内にHIF1αが誘導されたためなのかを検討中である。
