ヒト口腔扁平上皮癌におけるp27^<Kip1>発現と腫瘍分化及び予後との関連性

研究概要	ヒトp27^<Kip1>cDNA[Dr J Massague(Howard Hughes Medical Institute,Memorial Sloan-Kettering Cancer Center,NY)より供与]をpcDNA3.1(Stratagene)にセンス・オリエント並びにアンチセンス・オリエントに組み込んだ発現ベクターを作製し、ヒト口腔扁平上皮癌細胞株(B88)に導入してp27^<Kip1>遺伝子発現の制御を行い、p27^<Kip1>蛋白の過剰発現ならびに低発現が、B88細胞の増殖並びに腫瘍分化へ及ぼす影響につき検索したところ、in vitro並びにヌードマウスモデルにおいてp27^<Kip1>蛋白の過剰発現にて増殖能ならびに浸潤・転移能の低下が、p27^<Kip1>蛋白の低発現にて増殖能ならびに浸潤・転移能の増強が認められた。また細胞周期の解析にてp27^<Kip1>蛋白過剰発現株では、アポトーシスを示唆するsub-G1 peakの割合が増加していた。即ちヒト口腔扁平上皮癌におけるp27^<Kip1>蛋白の機能の一つとして、浸潤・転移抑制への関与が示唆されたため、今後、p27^<Kip1>蛋白の細胞接着因子並びに転移関連遺伝子の発現誘導との関連性につき検討が必要と考えられた。また腫瘍分化への影響については、HE染色像ではp27^<Kip1>蛋白の過剰発現にて明らかな変化が認められなかったが、低発現にて腫瘍の組織型が扁平上皮様から紡錘形を呈する細胞への変化が認められ、免疫組織染色像では、p27^<Kip1>蛋白の過剰発現にてIn volucrin並びにCAM5.2(Keratin7,8)の発現増強が認められた。これらの結果よりp27^<Kip1>蛋白の口腔扁平上皮癌終末分化との関連性が示唆された。
