疎水性分子表面積によるホスト・ゲスト複合体の構造と結合定数の予測

研究概要	まず、ホスト分子であるシクロデキストリンにゲストとして薬分子を取り込んで形成される複合体の構造と結合定数を予測するために、疎水性分子表面積を計算するソフトを開発した。計算された疎水性分子表面積の減少が最大になるときの複合体の構造が安定な構造であることおよびこの減少度と結合定数が相関することを示した。この相関性を利用して、任意のホスト・ゲスト系の結合定数をホスト分子とゲスト分子の構造から予測することが原理的には可能になった。一方、溶液中における薬(抗潰瘍薬と抗神経薬)や胆汁酸などの2量体およびシクロデキストリンと薬分子で形成される複合体の構造と結合定数をNMR法に基づいて決定した。このようにして実験的に決定された立体構造と疎水性表面積の定量的関係を研究しつつある。また、複合体の形成によって、薬の苦みや溶血性を抑制することができ、この抑制度を表面張力や起電力などの物性値を用いて、予測できることも実験的に証明した。その原埋は薬の苦みや溶血性は結合していない薬の濃度のみによって定まり、複合体はまったく苦みや溶血性を持たないことを利用している。苦みのような感覚試験は個人差があるが、このような物性値は再現性がよく、また、自動化できる可能性もある。以上の研究結果を総合すれば、最終的にはホストと薬分子の化学構造が与えられれば、薬の苦みや溶血性以外の生埋活性もコンピュータによる計算値あるいは物性の測定値から予測することが可能になる。この夢の実現のための確実な一歩を踏み出すことができた。
