細胞悪性化に関わるCD98シグナル伝達系の解析

研究概要

細胞悪性化に関わるCD98シグナル伝達系の解析を目的として、ヒトCD98hc cDNAとラットCD98hc cDNAを、各々、マウス線維芽細胞NIH3T3とBalb3T3にトランスフェクトし、細胞悪性化形質を調べた。また、CD98hcと結合する可能性のある新規アミノ酸トランスポーターのクローン化、新規CD98lcに対するモノクローナル抗体(mAb)の作製と対応抗原の性状解析を行った。
1、CD98hcの導入によりマウス線維芽細胞は軟寒天中コロニー形成、ヌードマウスでの腫瘍形成などの悪性化形質を示した。
2、悪性形質転換には導入CD98hcと内因性CD98lcとの結合が必要だった。
また、CD98hcの103番目のシステインがCD98lcとの結合に用いられていた。
3、CD98hcとの結合が示唆されているグルタミン酸受容体EAAC1を解析する過程で、欠失型EAAC1を同定、クローン化した。
4、新規CD98lcに対するmAbを作製し、この1cがhc同様、リンパ球活性化や癌化に伴って発現増強することを見出した。
5、CD98隣接分子としてアクチン結合性のトロポミオシンを同定した。
