研究概要	プロスタグランジン(PG)は生殖生理と深い関連が示されてきたが、どの受容体を介して、またどれくらい重要な働きを有するのかは全く不明であった。我々は、8種類のPG受容体欠損マウスを作製・解析し、PGが排卵(EP2)、分娩誘導(FP)、動脈管閉鎖(EP4)の各プロセスに必須の因子であることを見出した。本研究では、これらの実績をふまえ、(i)PGが各受容体を介して生理的に重要な機能を発揮する標的細胞を同定し、(ii)その細胞においてPGシグナルの有無により発現変化を呈する遺伝子群を探索することで、排卵・分娩・動脈管閉鎖の各過程のメカニズムを明らかにし、PG製剤の生殖領域応用の可能性を検索することを目的とする。平成11年度は、(1)まず排卵におけるEP2受容体について(i)の解析を行い、卵丘細胞に発現するEP2が必須であることを明らかにした(Hizaki et al.1999)。そこで現在、卵丘細胞での遺伝子発現変化の解析を行うために、野生型とEP2欠損マウスから卵丘細胞RNAを単離している。(2)また、分娩誘導におけるFP受容体の働きについては、黄体細胞におけるFPの働きがcriticalであることを見出しているが、FPの働きが黄体退縮を引き起こしてプロゲステロンの産生低下を来して子宮に分娩誘導を引き起こすことから、今回は、(ii)の解析を子宮に対して行うことで、FPシグナルの有無によって発現変化する遺伝子群の単離を行った。その結果、幾つかの発現変化遺伝子を単離し、これらの中にはFG合成酵素であるCOX-1、COX-2や、分娩への関与が示唆されるオキシトシン受容体の遺伝子が含まれていることを見出した(Tsuboi et al.2000)。さらに、他の単離遺伝子に関しては、妊娠後期の野生型マウス子宮における発現様式を解析し、分娩誘導に関与する可能性を検索している。
