高親和性カルニチントランスポーターの機能特性と薬物誘導型毒性発症機構との関連

研究概要	カルニチンは生体内脂肪酸代謝に必須な水溶性低分子物質であり、その組織移行はトランスポーターに大きく依存している。生体内カルニチン量の低下に伴うカルニチン欠損症は、最終的には心機能異常などの致命的症状の原因となり、また臨床的な一部の薬物投与により同様の症状発症が報告されており、薬物誘導型の二次性カルニチン欠乏症として問題となっている。研究代表者等は既に、カルニチンの腎での再吸収や組織分布に必須なカルニチントランスポーターOCTN2の遺伝子クローニングならびにその生理的役割について報告した。本研究ではOCTN2を介したカルニチンおよび薬物輸送機構を詳細に検討することにより、薬物によるOCTN2を介したカルニチン動態異常の可能性について明らかにすることを目的にした。ヒトOCTN2遺伝子をHEK293細胞に発現させ、その機能解析を進めた。その結果OCTN2を介したカルニチン輸送は種々の薬物によって阻害されることが示された。特に脂溶性の高いベラパミル、キニジンあるいはメピラミンなどによって強い阻害を受けた。またカルニチン欠乏症を引き起こすことが知られているバルプロ酸のような酸性薬物によっても阻害を受けた。また、これらカチオン性化合物は実際にOCTN2によって輸送されるが、その輸送機構はカルニチンとは異なりナトリウム依存性を示さないことがわかった。すなわちOCTN2はカルニチンとカチオン性薬物を異なるメカニズムで輸送する多機能性を有することが示唆され、新しいトランスポーター機能の概念を示唆するものとなった。今後は、そのメカニズムの詳細を解析するためにOCTN2安定発現細胞を作製し、阻害剤スクーリニングを簡便化するとともに、大量調整した細胞を基に膜小胞を調整し、輸送駆動力の解明や生理的輸送方向性を明らかにする予定です。
