接着分子をターゲットとした脳腫瘍細胞のアポトーシス誘導と治療への応用

研究概要

我々は脳腫瘍細胞において、酸化ストレス刺激で接着斑キナーゼ(FAK)からPI3-kinase-Aktサバイバル経路のシグナル伝達経路が活性化することを見い出し、すでにBBRC(1997)に報告した。今回はFAKの機能を明確にするために、wildFAKおよびmutantFAK3種をFAKを殆ど発現していない骨髄性白血病株HL-60にトランスフェクトし、その機能を調べた。mutant FAKとして増殖因子刺激で自己リン酸化する397,925のYをFにかえたもの、キナーゼ活性に重要な454KをRに変えたものを使用した。
その結果、wild-FAKを導入した細胞株は酸化ストレスおよび抗がん剤etoposideに対し抗アポトーシス機能を獲得したが、mutant FAK3種を導入した細胞株は抗アポトーシス作用を示さなかった。特にY397Fはコントロールより少し強く、細胞死を誘導した。これは既にJBC(2000)に報告した。従って、Y397Fはdominant negative FAKとして働く可能性があるので癌細胞で検討中である。また、FAK過剰発現株での抗アポトーシスのメカニズムをしらべたところ、FAK-PI3-kinase-Akt-NFkB活性化のサバイバル経路が活性化されていることがわかった。
