接着分子をターゲットとした脳腫瘍細胞のアポトーシス誘導と治療への応用

研究概要	我々は脳腫瘍細胞において、酸化ストレス刺激で接着斑キナーゼ(FAK)からPI3-kinase-Aktサバイバル経路のシグナル伝達経路が活性化することを見い出し、JBC(1999)に報告した。今回はFAKの機能を明確にするために、wild FAKおよびmutant FAK3種をFAKを殆ど発現していない骨髄性白血病細胞株HL-60にトランスフェクトし、その機能を調べた。その結果、wild FAKを導入した細胞株は酸化ストレスおよび抗がん剤etoposideに対し抗アポトーシス機能を獲得したが、mutant FAK3種を導入した細胞株は抗アポトーシス機能を示さなかった。むしろY397F変異株はアポトーシスを誘導する傾向を示した(JBC,2000)。従って、Y397FがFAKに対しdominant negative的に働いている可能性があるので脳腫瘍細胞にアデノウィルスベクターを用いて導入したところ、アポトーシスを誘導することを見い出した。またFAK過剰発現細胞株での抗アポトーシスのメカニズムをしらべたところ、FAK-PI3-kinase-Akt-NFκBのサバイバル経路が活性化されていることがわかった。また、このサバイバル経路を遮断すれば効率よく癌細胞にアポトーシスを誘導できると考えられるので、その薬物を検索した。その結果、免疫抑制剤として開発中の薬物FTY720がこのサバイバル経路を遮断し、癌細胞にアポトーシスを誘導できることを見い出した(BBRC,2001)。
