動物レクチンと糖鎖間の多価特異的相互作用の生物学的意義の解明

研究概要

今日知られているレクチンのほとんどは複数の糖鎖結合部位を含み、複合糖質と多価の相互作用を行う。ここに生体制御タンパク質としてのレクチンを理解するための重要な鍵が隠されているので、動物の代表的レクチンであるガレクチンを主たる対象として取りあげ、多価特異的相互作用の原理の解析と解明を目指した。この目的には新しい有効な研究手段の開発が必須なので、高感度、迅速、精密な相互作用研究法としてわれわれがこれまでに発展させてきたフロンタルアフィニテイークロマトグラフィー法、キャピラリー電気泳動法について、更に強力なものとするために改良を重ねた。これを用いて哺乳動物、線虫、カイメン等の各種動物由来のガレクチンをリコンビナントタンパク質として用意し、数十種類にのぼるさまざまな構造を持つ糖鎖との解離定数を系統的に調べ、各ガレクチンの糖結合特異性に関するプロファイルを作成した。この結果は、ガレクチンの細胞内外での機能、分子進化、その他多彩な点に関して多くの示唆を与えるものとなった。多価の相互作用に関しても多くの知見が得られ、ガレクチンが細胞表面等において集合体を形成することにより、多価の複合糖質との相互作用が劇的に影響を受ける可能性が示された。
