GCP364およびGCP170に相互作用を持つゴルジ装置タンパク質の検索

研究概要

ゴルジ装置局在II型膜蛋白質GCP364/giantinとゴルジ装置ペリフェラル膜蛋白質GCP170それぞれの局在化に必要なドメインを用い、酵母のTwo-Hybrid系で相互作用を持つタンパク質の検索を行った。
1)GCP364は細胞質C末の2938-3116アミノ酸の領域が強いゴルジ局在能を示した。ここをbaitに用いてHeLa細胞cDNA libraryをscreeningした結果、新規ゴルジ局在タンパク質GCP60がGCP364のゴルジ局在領域と相互作用を示した。GCP60はゴルジ装置細胞質側に存在するペリフェラルな膜タンパク質であった。免疫電顕による観察ではゴルジ装置cisternaに存在しており、GCP364と同じ分布をしめした。in vitro結合実験でもGCP364とGCP60の結合は特異的であった。GCP60を過剰発現したCOS-1細胞でゴルジ装置が崩壊し、小胞体からゴルジ装置への輸送が止まることから、GCP60はGCP364と共にゴルジ装置形成を担うと考えられる。
2)GCP170はそのN-末から約400アミノ酸の領域がゴルジ局在に大きな働きをしている。ゴルジ装置局在化において、必要な最小のアミノ酸配列を検討した結果、159-237アミノ酸の領域が強力なゴルジ局在能をしめした。この領域をbaitにしてTwo-Hybrid Screeningをおこなった結果、GCP170と相互作用を持つ数種類の未知のゴルジ装置タンパク質が検出された。これらのタンパク質の性質、細胞内局在について検討中である。
