研究概要	RNAポリメラーゼのσサブユニットは、RNAポリメラーゼによるプロモーターの認識と、転写開始点付近のDNA二重鎖の開裂に中心的な役割を果たすと考えられている。大腸菌における主要σであるσ70、終止期特異的に働くσ38、窒素代謝関連遺伝子の発現に働くσ54の3種のσ因子を対象に、分子上の様々な位置に1個のみシステイン残基を持つ変異体をそれぞれ12種類、9種類、5種類作成した。システイン特異的な修飾反応を使って、これらのσ変異体のそれぞれにペプチド鎖・ヌクレオチド鎖切断活性を持つ鉄・EDTA誘導体を導入した。ホロ酵素を再構成したのち、切断反応を行なって、RNAポリメラーゼのβ、β'サブユニット上の切断位置を同定した。また、DNAを加えて転写開始複合体を形成させたのちに切断反応を行ない、鋳型DNAの切断位置を調べた。その結果から、3種類のσ因子がコア酵素上のほぼ同一の領域と接触していること、転写開始複合体中でのσ因子の配向が互いによく似ていることが示された。特にσ54は一次構造上は他のσ因子と全く似ていないにもかかわらず、高次構造およびコア酵素やプロモーターDNAとの結合様式が他のσと類似していることを示唆している点で興味深い。 σ70の保存領域のうち機能がわかっていない4.1領域に注目し、保存性の高いアミノ酸をAlaに置換した変異体11種を作成して、in vitro転写およびDNaseIフットプリンティングによる解析を行なった。また、529番め以降のアミノ酸配列を持つC末端フラグメントに同じ11種類の変異を導入して、N末端フラグメントとの相補性に与える影響を調べた。それらの結果から、4.1領域はプロモーターの認識に直接関わってはいないものの、4.1領域上のいくつかのアミノ酸残基が、σの4.2領域とプロモーターの-35配列との正確な配向を決めるのに重要な役割を果たしていることが示唆された。
