研究概要	種々のサイトカインは、その蛋白分子の獲得に端を発し、サイトカインの機能の解明に向けて多大な研究がなされ、研究はレセプターから核へ向かうシグナル伝達の解析へと進んだ。近年、サイトカインレセプターに会合するチロシンキナーゼとしてJakファミリーキナーゼの存在が明らかにされると、さらに、このJakファミリーキナーゼの主要な基質蛋白としてSTAT蛋白が同定された。申請者はサイトカインシグナル伝達系のJakファミリーキナーゼの制御に重要なJakファミリーキナーゼインヒビターであるSOCS-1とJakファミリーキナーゼとの結合部位も同定した。また、JakファミリーキナーゼのうちTyk2に対するノックアウトマウスを作成し、このマウスにおいてはサイトカインのうち、インターロイキン12のシグナル伝達系が障害されることを示した。現在、このノックアウトマウスを用いることによりインターロイキン12レセプターの下流に位置する新たな遺伝子、あるいは蛋白の同定を試みている。申請者はさらにサイトカインのシグナル伝達系の再構築系を作成し、これを用いることにより種々のサイトカインシグナル伝達系と他のシグナル系のクロストークの存在を分子レベルで明らかにした。特にサイトカインとステロイドレセプターのシグナル伝達系のシグナルトークについてより詳細な分子機構の解析を進めている。
