リンパ球における接着分子セレクチンの二種のリガンドの発現機構の研究

研究概要	リンパ球は、炎症反応の際に炎症局所の病巣に動員されるだけでなく、日常とくに病変のない場合にも、末梢血管から遊走して皮膚や体腔中を徘徊し、やがてリンパ管を介して血中に戻るという平常時の組織へのホーミングを示す。最近我々は、リンパ系組織において、これまでにセレクチンのリガンドとしてすでに良く知られたシアリルLe^Xに加え、シアリル6-スルホLe^Xをセレクチンの新しいリガンドとして同定した。これにより、リンパ系組織にはシアリルLe^Xとシアリル6-スルホLe^Xの二種のセレクチンリガンドが存在することとなった。本研究の目的は、これらのリンパ球の二種のセレクチンリガンドの生理的意義および機能分担と、それぞれの発現調節機構を検討した。その結果、炎症反応の際の動員においてはシアリルLe^Xが接着分子セレクチンのリガンドとして機能し、平常時の組織へのホーミングにおいてはシアリル6-スルホLe^Xがセレクチンのリガンドとして機能すると見られる成績を得た。平常時のリンパ球のホーミングにおいても、炎症時の動員においても、血管外脱出の過程はまずリンパ球と血管内皮細胞との接着にはじまる。この時、セレクチンと対応する糖鎖リガンドとを介した細胞接着が重要な役割を演じる。リンパ球の日常的ホーミングにおいては硫酸化糖鎖シアリル6-スルホLe^Xがリガンドとして機能し、SLC-CCR7,TARC-CCR4などのホメオスタティック・ケモカイン系との協同作用によってホーミングが起こる。一方、炎症においては、セレクチンリガンドのうち硫酸基を持たないシアリルLe^Xの発現がリンパ球上に強く誘導され、これによる細胞接着と炎症性ケモカインの協同作用によって炎症性細胞浸潤が起こる。このように、生体は炎症時と健常時とでセレクチンの二種のリガンドを使い分けており、炎症刺激にともなってリンパ球上のリガンドの発現が交替する事が判明した。
