ヒトAPP導入マウスを用いたアミロイド蛋白神経毒性の神経病理学的研究

研究概要	高齢化社会で重要課題であるアルツハイマー病研究を多くの研究者層によって協力的に推進するために、また安定した実験系を構築するためにもモデル実験系としての細胞培養系と病態モデルマウスの作成は重要な意味を持つ。われわれはプリオンプロモーターによる制御下に予定遺伝子を組み込んだ国産のトランスジェニックマウスの作成を推進している。まずアルツハイマー病に(1)特異的な老人斑形成マウスを作成すべく変異型APPと変異型PS1のマウスの2種類を作成し、両者の掛け合わせにより、アルツハイマー病モデルマウスを作成する戦略をたてている。ここでいう変異型は早期発症型家族性アルツハイマー病で見いだされた遺伝変異であり、前者はAPP670/671(スウェーデン変異)とAPP717(ロンドン変異)の組み合わせ(NLF変異と呼称する)であり、後者はPS1A260Eである。遺伝変異はPCRにより人工的合成により導入したものを元となるヒト遺伝子に組み込み、全塩基配列を確認した後、マウス初期胚に微量注入した。これによって作成されたマウスは各々単独で老人斑を形成するには著しく年月がかかるものの、両者の掛け合わせにより、ヒトの50年をマウスの数ヶ月に短縮することが出来ると報告されている。一方(2)同時に並行して本疾病に特徴的な神経病変である神経原線維変化形成マウスも進行中である。このためにV337M変異型タウをもつマウスを作成したが、8ヶ月現在で病変が現れていない。同時に、神経毒性の研究を胎児から調製した海馬ニューロンを用いて実行しつつあり、すでに合成ペプチドを用いた実験系で既報値より鋭敏な神経毒性の探索が可能な段階を達成している。
