パーキンソン病原因遺伝子NACP/α-シヌクレインによる神経変性機構

研究概要

筆者らがAlzheimer病(AD)脳アミロイドの分析から未知分子としてクローニングしたNACP(non-Aβ component of AD amyloid precursor)は、α-synucleinとも称され、その遺伝子変異が家族性Parkinson病(PD)の疾患原因遺伝子として同定された。我々はPDの特徴的神経病理であるLewy小体の本態がNACP全蛋白質であることを示した。Lewy小体はPDのみならず、痴呆が主症状のLewy小体型痴呆(dementia with Lewy bodies:DLB)の本態であり、Lewy小体病として統括される。又、従来その本態が不明であった多系統萎縮症(multiple system atrophy:MSA)の細胞内異常繊維構造がNACPであることも我々は示した。従って、NACPはこれらの神経変性疾患発症に共通した鍵分子であると考えられ、その分子病理の解明を主目的にしている。
ラットPC12細胞は、NGF添加により神経細胞に分化する事が知られている。この細胞がNACP/α-synucleinファミリーのどのタイプを発現しているかを知るために、RT-PCRで解析した結果、β-synuclein(PNP14)のみを発現している事を前年度報告した。これまでLewy小体等の病理的構造体にβ-synucleinが検出された報告はなく、NACPのみが異常蓄積すると考えられている。この細胞にNACP発現ベクターを導入して得られた40クローンをWestern blotで解析した。蛋白質の検出は独自に開発した特異抗体(MDV2、EQV1、PQE3、REE1)を用いた。その結果4クローンのみに発現が確認された。次に、PDの動物モデル作成の際に汎用される神経毒であるMPP^+、OHDA、BSO等で細胞を処理してvulnerabilityを検討したところ、BSO存在下でNACP発現細胞はより死にやすくなっていた。
