研究概要

家禽雛鳥の認知機能を明らかにすると共に、記憶形成と行動制御の脳内メカニズムを解析するために一連の解析を総合的に展開した。個体レベルの行動学的解析・脳スライス標本を用いた神経生理学的解析・最初期遺伝子発現を指標とした分子レベルの解析・課題遂行中の覚醒自由行動下における大脳単一ニューロン活動の解析を、総合的に行なうことにより、以下の知見を得た。
(1)行動学的研究:新規学習課題の開発と記銘内容の解析
受動的回避学習と馴化学習を指標として雛鳥の想起の内容を解析したところ、一義的には対象の色を詳細に記銘し想起していることが判明した。他方、対象の形に対する弁別カテゴリーは単純で、円形の輪郭を持つものに限局していた。さらに、水を報酬とするGO弁別課題・餌を報酬とするGO/NO-GO弁別課題を開発しその記銘内容を解析したところ、色を第一義として位置を第二義とする多元的な記憶を形成していることが判明した。
(2)記憶の素過程に関する生理学的研究:シナプス長期増強と最初期遺伝子発現
大脳IMHV領域(哺乳類の皮質連合野に相当)のスライス標本において、局所興奮性シナプスの長期増強と最初期遺伝子発現の間の因果的関連を吟味したが、両者は基本的に独立であることを示唆する結果を得た。さらに、基底核LPO領域において、D1受容体依存のシナプス長期増強とNMDA受容体依存の長期抑圧が同一ニューロンに同時に生起する現象を見出した。
(3)記憶の細胞表現に関するシステム生理学的研究:記憶に基づく予期と注意の符号化
自由歩行下のIMHV領域・LPO領域より単一ニューロン活動を導出し、GO弁別課題・GO/NO-GO弁別課題遂行中の関連活動を解析した。IMHV領域からは視覚・聴覚性注意を表現するニューロンが、LPO領域からは報酬評価と報酬予期を表現するニューロンが、それぞれ見出された。さらに、LPO領域のカイニン酸局所破壊は水を報酬とするGO弁別課題の成立を著しく阻害したが、一度形成された弁別課題の遂行に影響を与えず、刷り込み記憶の形成・発現ともにも影響を与えなかった。
