研究概要	本研究は、神経伝達物質合成酵素の活性の変化が神経伝達物質の合成量の変化を通してどのように神経機能の変化を起こしうるかということを調べることを目的としている。チロシン水酸化酵素はカテコールアミン生合成の律速酵素であり、カテコールアミンの合成量を調節している。そこで、チロシン水酸化酵素遺伝子のイントロン内にCre組換え酵素の認識配列であるloxPを相同組換えにより導入したマウスと、Creを発現するマウスとを掛け合わせることにより組織特異的および時期特異的遺伝子破壊を実現する。この時、Creはエストロゲン受容体のリガンド結合部位との融合タンパク質として発現させて、リガンドの投与によりCreの組換え酵素活性を誘導できるようなもの(Cre-ERT)を用いる。これまでに、チロシン水酸化酵素のイントロン内にloxP配列を導入したマウスを作製した。また、Cre-ERTをチロシン水酸化酵素のプロモータの制御下に発現するマウスを作製するために、Cre-ERTをチロシン水酸化酵素の遺伝子内に導入したマウス(Cre-ERTノックインマウス)を作製する。これまでに、Cre-ERTノックインマウス作製のためのターゲッティングベクターの作製とES細胞での相同組換え体の単離に成功し、生殖細胞系列にCre-ERTをもつマウスを検索中である。今後、loxPを導入したマウスとCre-ERT発現マウスを掛け合わせることにより、誘導的破壊実験を行う予定である。
