糸状菌における転写制御因子CCAAT結合複合体の解析

研究概要	MJ Hynes教授のグループはA.Nidulans のamdS遺伝子を中心に、窒素および炭素化合物の利用におけるamdS遺伝子の発現を制御する遺伝子に関して、膨大な遺伝学的成果を蓄積している。また、糸状菌の遺伝子操作も世界に先駆けて開始しており、糸状菌の分子生物学に関する多くの経験を積んでいる。一方、我々はタカアミラーゼA遺伝子の転写制御エレメント及び転写制御因子の解析からCCAAT配列及びHAP複合体がタカアミラーゼA遺伝子の転写促進に重要な役割を演じており、amdS遺伝子上のCCAAT配列と共通した機能を示すことを明らかにした。本共同研究においてはCCAAT結合複合体に関してin vitro,in vivoの両面から解析することを計画した。共同研究を計画した時点では、我々のグループはA.OryzaeにもCCAAT結合因子、AoCP、が存在することを確認していたので、AoCPを構成するサブユニットの遺伝子を分離することから共同研究を開始した。Hynes教授のグループから分与されたhapC遺伝子を使用して麹菌よりAohapC遺伝子をクローン化し、遺伝子構造を明らかにした。現在はすべてのサブユニット遺伝子を麹菌より取得し、構造解析も完了している。一方、CCAAT結合複合体のin vivoにおける機能解析を目的として、Hynes教授のグループは遺伝学的にhapC遺伝子が欠損した株を取得した。また、我々はAohapC遺伝子をこの変異株へ形質導入したところ、HAPCの機能がAoHAPCで相補されることを発見した。
