香港で発生した新型H5インフルエンザウイルスの起源と変異

研究概要	1997-1998年にかけて香港で6名の市民を死に至らしめた鶏由来家禽ペスト型インフルエンザウイルスは鳥由来ウイルスでありながら哺乳動物に対しでも病原性を示すウイルスでった。一方、我々はこれまで野生水禽由来の弱毒インフルエンザウイルス(499株)を鶏で継代することにより、弱毒株が強毒の家禽ペストウイルスに変異することを明らかにしてきた。その過程で本ウイルスが香港ウイルスと同様に哺乳動物であるマウスに対しても病原性を獲得していることを明らかにした。本研究の最終年度である本年度はまずニワトリに対して増殖性の異なるこれら一連の継代株の遺伝子を詳細に比較解析した。さらにその解析結果をもとに種々の変異を導入した人工ウイルスをリバースジェネティクスにより作出してその性状を調べることにより、インフルエンザウイルスの宿主適応や病原性獲得機序の解明を目指した。その第一段階としでまずリバースジェネティクスによる組み換え499(r499)株を作出した。親株及びそのニワトリ継代株(24a)の8本のウイルス遺伝子分節の全塩基配列を決定し、両株間での推定アミノ酸配列の比較を行った結果、PB2蛋白3ケ所、PB1蛋白1ケ所、PA蛋白1ケ所、NP蛋白2ケ所、HA蛋白13ケ所、NA蛋白4ケ所、M1蛋白2ケ所(M2蛋白1ケ所及びNS1蛋白2ケ所のアミノ酸置換と24a株におけるNS1の218-247aa領域の欠損が確認された。次に親株の各分節遺伝子cDNAをpol.ベクターにサブクローニングし、各々のプラスミドをトランスフェクションした293T細胞の培養上清を発育鶏卵尿膜腔内に接種したところ感染価23×10^7PFU/ml、HA価128倍のr499株を回収することに成功した。以上の成績から、リバースジェネティクスにより水禽由来インフルエンザAウイルス499株の組み換えウイルス系が確立された。現在この実験系を用いて、最終段階である感染実験による解析を計画している。
