ポジトロン標識合成法の新展開と3D-PETによる新しいヒト機能検査法の開発

研究概要	1.ポジトロン標識化合物の開発。 ^<11>C-ヨウ化メチルと同様に汎用性のあるポジトロン標識前駆体である^<18>F-標識ヨウ化フルオロベンジル反応を確立した。標識合成の迅速で効率的な^<18>F-標識フルオロベンジルイミダゾイルプロピルエーテル(^<18>F-FUB193、^<18>F-fluoropropoxyfan)の反応条件を確立し、結合実験よりその特異性を調べた。また^<11>C-ヨウ化メチルを合成する方法として、新しくガス相法を確立して臨床用の^<11>C-リガンドを用いた臨床研究に応用した。また^<11>C-ヨウ化メチル法より合成収率の高い^<11>C-メチルトリフレート法を確立して、新規標識薬剤として^<11>C-ドネペジルを合成した。 2.3次元PETカメラを用いたヒト脳の神経伝達量測定法の開発と疾患への応用。 3次元PETカメラの臨床応用としてアルツハイマー型痴呆患者においてヒスタミンH1受容体量を測定し、アルツハイマー病の認知機能低下に伴ってH1受容体が減少することが分かった。また現在、うつ病、精神分裂病、ナルコレプシーにおいてH1受容体を測定している。 3.脳賦活試験法による高次脳機能の研究。 (1)脳賦活法を用いたヒスタミン神経系の研究。本研究は、まず(1)[^<11>C]ドキセピンを用いてH1受容体占拠率を測定し、眠気発生や認知能力低下にどれだけのH1受容体占拠が必要か明らかにすることを試みた。(2)H_2^<15>Oと3D-PETを用いて視覚認知課題遂行時の脳機能イメージングを行い、抗ヒスタミン薬投与による眠気や認知機能低下の発生メカニズムを研究した。(3)情動を測定するために顔の認知タスクの開発と評価を行い、H_2^<15>Oと3D-PETを用いて情動のメカニズム研究を行った。
