研究概要	結核菌をはじめとするMycobacteriumは肉芽腫を形成することが知られているが、その機構は不明である。パスツール研究所のG.Gachelin博士のグループとの共同研究でMycobacteriumによって誘導される肉芽腫の形成過程を詳細に解析した結果、肉芽腫形成の初期にNKT細胞まず局所に集まることが明らかとなった。NKT細胞を欠くJα281ノックアウトマウスでは肉芽腫の形成が著明に減少することなどから、肉芽腫の形成にはNKT細胞が重要な役割を持つことを明らかにした。 LFA-1は白血球の細胞間相互作用に必須の役割を持つインテグリンファミリーの接着分子であるが、その機能的な重要性にもかかわらず細胞内への信号伝達経路に関しては全く不明である。米国のDNAX研究所のL.Lanier博士らとの共同研究で、最近新しくクローニングされた接着分子であるDNAM-1がLFA-1からの信号伝達に重要な役割を持つことを明らかにした。リンパ球上においてT細胞受容体からの信号はLFA-1分子にinside-outのシグナルを与え、リガンドとの結合性が上昇すると同時にDNAM-1と会合する。LFA-1分子がリガンドと結合するとsrcファミリーのチロシンキナーゼであるFynが結合することによりDNAM-1に燐酸化が起こり細胞内に信号が伝達されることを明らかにすることができた。
