真核細胞のDNA複製とチェックポイント制御の分子機構に関する共同研究

研究概要	分裂酵母とヒトのCdc7類似キナーゼ複合体である、Hsk1/Him1およびhuCdc7/H37の転写とタンパク質の細胞周期における変動を検討した結果、いずれの生物種においても、触媒サブユニットは細胞周期を通じてほぼ一定のレベルで発現されること、制御サブユニットの発現はG1期で低下しG1後期からS期にかけて上昇しS期を通じて高く保たれることが示された。 muCdc7遺伝子の2つのalleleを欠損したマウスは胎生3.5日から6.5日の間に死亡した。さらに、muCdc7遺伝子の不活性化を誘導できる変異E S株を作製し、muCdc7の機能が動物細胞のDNA複製に必須であることを証明した。一方ヒトCdc7/H37複合体を精製し、in vitroリン酸化反応による解析を行った結果、MCM複合体の中のMCM2サブユニットが特異的にリン酸化されin vivoで観察されるリン酸化パターンと類似したリン酸化パターンが得られた。また、MCM2上のリン酸化部位の詳細な解析から重要なリン酸化部位をほぼ同定し、Cdc7はMCM2のリン酸化を通じてその複合体の組成を変化することによりヘリカーゼ活性を制御する可能性を指摘した。 competitivePCR法を用いてヒト5番染色体上GMCSF-IL3領域(5q)に複製起点が存在することを示した。興味深いことに、その複製起点の活性化のパターンはT細胞と、サイトカイン遺伝子発現を行っていない非T細胞においてて異なっている可能性が示唆された。
